چکیده

Abstract Background Ureaplasma spp . are associated with various infectious diseases in females, but there is still limited evidence regarding whether they related to nonspecific cervicitis. The aim of this study was develop and evaluate a digital droplet PCR (ddPCR) assay for the detection quantification spp. cervical swabs. Methods A total 267 non-specific cervicitis (NSC) patients 195 asymptomatic females were included study. We produced standard curves analytical performance ddPCR assay. Then, we detected quantified bacterial load Results prevalences U. parvum 37.8% (101/267) 29.7% (58/195), urealyticum 9.0% (24/267) 8.7% (17/195) NSC group control group, respectively. In addition, median copy number 2.5 × 10 4 copies/ml ( n = 101) 9.2 3 58) group. significantly higher than that individuals P < 0.001). whereas 8.4 24) 1.4 17) two groups, respectively, , difference not statistically significant 0.450). Conclusions Our first method clinical samples, has excellent wide range application prospects.

Sensitive and accurate quantification of human malaria parasites using droplet digital PCR (ddPCR)

Accurate quantification of parasite density in the human host is essential for understanding the biology and pathology of malaria. Semi-quantitative molecular methods are widely applied, but the need for an external standard curve makes it difficult to compare parasite density estimates across studies. Droplet digital PCR (ddPCR) allows direct quantification without the need for a standard curv...

ddpcr: an R package and web application for analysis of droplet digital PCR data

Droplet digital polymerase chain reaction (ddPCR) is a novel platform for exact quantification of DNA which holds great promise in clinical diagnostics. It is increasingly popular due to its digital nature, which provides more accurate quantification and higher sensitivity than traditional real-time PCR. However, clinical adoption has been slowed in part by the lack of software tools available ...

Diagnostic utility of droplet digital PCR for HIV reservoir quantification

Quantitative real-time PCR (qPCR) is implemented in many molecular laboratories worldwide for the quantification of viral nucleic acids. However, over the last two decades, there has been renewed interest in the concept of digital PCR (dPCR) as this platform offers direct quantification without the need for standard curves, a simplified workflow and the possibility to extend the current detecti...

Comparison of next-generation droplet digital PCR (ddPCR) with quantitative PCR (qPCR) for enumeration of Cryptosporidium oocysts in faecal samples.

Clinical microbiology laboratories rely on quantitative PCR for its speed, sensitivity, specificity and ease-of-use. However, quantitative PCR quantitation requires the use of a standard curve or normalisation to reference genes. Droplet digital PCR provides absolute quantitation without the need for calibration curves. A comparison between droplet digital PCR and quantitative PCR-based analyse...

Transgene Detection by Digital Droplet PCR

Somatic gene therapy is a promising tool for the treatment of severe diseases. Because of its abuse potential for performance enhancement in sports, the World Anti-Doping Agency (WADA) included the term 'gene doping' in the official list of banned substances and methods in 2004. Several nested PCR or qPCR-based strategies have been proposed that aim at detecting long-term presence of transgene ...
